Jimenez-Moreno et al. (bioRxiv, Nov 2025) identify a new, PKA-independent arm of the cAMP pathway: cytoplasmic TurboID proximity proteomics across five ER-phagy receptors ranked PRKAR1A as the strongest activator of FAM134B/C flux, and CRISPR knockout of Prkar1a suppressed basal (Atg7-dependent) FAM134B/C flux while forskolin (FSK) still activated flux in control cells but was blunted by Prkar1a loss β ruling out canonical PKACAT suppression as the mechanism . Molecularly, an amphipathic helix at FAM134B residues 385-421 (mapped to 392-408; I400/I417 critical) binds a hydrophobic pocket in the PRKAR1A N-terminal dimerization domain, supported by recombinant in vitro binding, AlphaFold3 modelling and a FoldX ΞΞG of 1.69 kcal/mol for the I400R-disrupting mutation .
FRAP shows PRKAR1A in FSK-induced foci recovers rapidly (liquid-like) while FAM134B-EGFP does not (membrane-immobilised); SoRA and CLEM resolve these as irregular, partially non-membrane-delimited condensates making extensive ER contacts with embedded LAMP1+ lysosomes β framed as "degradation hubs" . Condensate disruption (1,6-hexanediol, 2.5%) or ΞIDR PRKAR1A ablates FAM134B-LC3B clustering, while mtPS (PKACAT-binding deficient) PRKAR1A rescues it, consolidating kinase independence .
FSK-shifted FAM134B proximity proteomes enrich Rho-family Reactome signatures; FAM134B/C co-IP binds inactive RhoA (T19N) selectively over active G14V, and T19N RhoA is preferentially trafficked to lysosomes in an FSK-, Fam134b/c- and Prkar1a-dependent manner; downstream P-MLC2 suppression is reversed by Y27632 in knockouts . Physiologically, the axis drives cAMP-dependent protrusion, reduced collective wound closure, and a collective-to-single-cell invasion switch in 3D collagen spheroids (rescue requires I400 and LIR); epinephrine reproduces the switch . This complements known condensate biology of cAMP-bound PRKAR1A and connects to parallel findings that ER-phagy receptors regulate non-ER-proteostasis clients (e.g., SERCA1 in statin myopathy ).
Schematic network of the sourced mechanism: cAMP β PRKAR1A condensates β FAM134B/C clustering with LC3B β lysosomal degradation hub co-sequestering RhoA β reduced ROCK/P-MLC2 β morphology/invasion switch (per the preprint and Cell 2020 condensate work).
The authors themselves flag overexpression of tagged baits, non-specific 1,6-hexanediol, and the largely correlative link between condensates and flux . Additional BGPT-noted caveats: almost all mechanistics use one murine KPC PDAC line (HEK293FT only for co-IP), so cell-type generality is asserted, not shown; RhoA itself was not confidently quantifiable in the proteome β the RhoA connection rests on pathway enrichment of its interactors plus co-IP; an alternative explanation (RhoA as indirect passenger of lysosome-delivered membranes) is not formally excluded. Replication at endogenous expression levels, a second cancer type, or in vivo metastasis data would falsify or strengthen the model.
