Working on the Great Barrier Reef (Swain Reefs, July 1981), the authors isolated zooxanthellae from a single head of Stylophora pistillata over 5–6 days and counted blebbing fat droplets by phase-contrast microscopy within 5 min of removal from the sea . Key quantitative findings: blebbing cells peaked at 11–13% between 14:00–19:00 and fell to 5–7% in early morning; division averaged 2% (4% early morning); ~1.7 ng lipid per cell (~7.4% of the 23 ng cell mass) .
Note: intermediate points interpolate the reported min/max ranges; only endpoints are direct observations.
¹⁴C-acetate incorporation showed triglyceride as the dominant algal storage lipid with no wax ester synthesis, consistent with host wax esters being modified downstream . Oxygen bubbling stimulated lipid synthesis (167.4 vs 96.6 nmol acetate mg Chl⁻¹ h⁻¹ under O₂ vs air) while depressing photosynthesis, hinting at photorespiratory carbon routing . This builds on earlier work showing most coral lipid synthesis occurs in zooxanthellae while most lipid resides in host tissue .
Strengths: in-situ sampling, prompt microscopy (<5 min), isotope tracing, and consistency across species (soft corals Sarcophyton, Lobophyton also blebbed; Tridacna and Millepora did not) .
Weaknesses: extrusion was never directly observed—cells failed to extrude on slides within 2 h and some lysed; counts are underestimates (back sides invisible); no replicates beyond n=2 preparations/time point; no statistical testing; the O₂-stimulates-lipogenesis mechanism remains speculative; and the N₂>air lipid synthesis anomaly is unexplained . The triglyceride-to-wax-ester conversion hypothesis is presented without experimental support. The finding is nevertheless corroborated by the later demonstration that coral lectins bind zooxanthellae in a lipid/membrane-dependent manner .
Modern surveys confirm zooxanthellate corals are lipid-rich (hexacoral triacylglycerols ~23.9% of lipid classes) , and lipid mediation of symbiosis is now recognized more broadly through octadecanoid signaling .
