The study addresses a previously contested question: is the 45S particle that accumulates in E. coli ΞrlmE a dead-end degradation product, or a real 50S assembly precursor? The authors resolve this with three convergent lines of evidence. First, pulse-chase ΒΉΒ³C-adenosine labeling shows ΒΉΒ³C-rRNA in 50S increases in the first 40 min while ΒΉΒ³C in 45S plunges, inconsistent with 45S being a breakdown product . Second, rifampicin transcription shutoff shows ~half of 45S converts to 50S within 120 min. Third, the 45S particle lacks mΒ³Ξ¨1915 β a 70S-specific modification deposited by RlmH β ruling out a mature-50S origin .
The mechanistic core is elegant: recombinant RlmE + AdoMet converts ~20β25% of 45S to 50S in vitro β the first demonstration of enzymatic 50S formation from a precursor by an assembly factor β and the reconstituted 50S productively joins 30S to form 70S. Critically, the U2552C mutant, which still binds RlmE (SILAC shows equal occupancy) but loses ~85% of Um2552 methylation, still accumulates 45S. This cleanly separates enzyme binding from modification function, showing the modification itself, not RlmE binding, triggers assembly . The mechanistic model β Um2552 stabilizes the Um2552βC2556βU1955 base-triple, promoting H92βH71 interdomain contact, L36 recruitment, central-protuberance positioning β is supported by hydroxyl-radical probing showing 45S-specific cleavages exactly in H92/H71 and the L36 pocket (H89/H91/H97/H42) . The rlmEβrpmJ synthetic lethality (double mutant cannot grow at low temperature) and absence of L36 from 45S independently corroborate cooperativity between Um2552 and L36.
The central precursor claim is well-supported (isotope tracing + rifampicin chase + mΒ³Ξ¨1915 absence) and the modification-vs-enzyme dissection is a genuine strength. The conclusion would be undermined if (a) purified 45S converted to 50S without Um2552 formation, or (b) direct peptidyl-transferase assays showed the reconstituted 50S nonfunctional. High confidence in the E. coli findings; moderate confidence in the atomic-level mechanism, which is model-based inference from footprinting rather than direct structure determination of the 45S precursor.
