The paper (Communications Biology, 2024; doi:10.1038/s42003-024-07437-2) proposes: HFD β NF-ΞΊB recruits Brd4 to a shared Ccr2/Ccr5 enhancer β eRNA/mRNA synthesis β colonic macrophage infiltration β dysbiosis and barrier leak β LPS (metabolic endotoxemia) β obesity and insulin resistance . The strongest links are in vitro ChIP/eRNA knockdown (n=3 each) showing Brd4, RNAPII, and RelA occupancy at the enhancer and siRNA eRNA depletion reducing Ccr2/Ccr5 and MCP-1 migration. In vivo links rest on Brd4-CKO vs WT comparisons (n=4β7 per group) at 4 and 20 weeks HFD, plus a LPS-minipump rescue experiment .
Reported observations vs interpretation. Reduced F4/80+ cells, lower Ccr2/Ccr5, restored Akkermansia muciniphila, Bacteroides acidifaciens, and Butyricicoccus pullicaecorum, lower plasma FD4 and LPS are observations. The causal ordering (colon-first, then adipose) is author interpretation; eRNA causality relies on siRNA knockdown, which has off-target risks not ruled out by rescue with eRNA refills. The key falsification result β LPS infusion restoring phenotype β supports the endotoxemia arm, but only in Brd4-CKO mice, not WT controls infused with LPS in parallel .
