Targeted allelic-exchange deletion of csxA in C. sporogenes NCIMB 701792 produced spores that completely lacked the balloon-like exosporium while the cortex, coat, interspace and parasporal layers remained intact (WT n=73, ΔcsxA n=17 by negative stain; WT n=32, ΔcsxA n=34 by thin section) . Conversely, ΔcsxC spores retained the exosporium but lost all parasporal layers (ΔcsxC n=8 negative stain; n=27 thin sections), genetically separating the two assemblies .
CryoEM 3D electron crystallography of recombinant CsxA crystals resolved structure factors to 9 Å in-plane and 20 Å perpendicular, revealing hexagonally arranged crown-like assemblies interconnected by trimeric linkers; AlphaFold3 predicts CsxA as a cysteine-rich SPOCS/DUF3794-domain trimer fitting this density .
Parasporal layer sheets imaged by AFM showed a 63 ± 6 Å lattice (n=31) with screw dislocations and point defects, layers stacked in register with 50–70 Å step heights (n=18), and lattice continuously resolved around dislocation cores—direct evidence that a native proteinaceous assembly grows via screw-dislocation-mediated crystallisation, previously demonstrated mainly in inorganic crystals and biomineralised nacre . Notably, the exosporium contributes little to dehydrated spore dimensions, while ΔcsxC spores show reduced dehydrated volumes, implying the parasporal layers contribute substantially to envelope bulk.
The authors concede several gaps: CsxC is not directly localised within the parasporal lattice (assignment rests on the ΔcsxC phenotype, mass-spectrometry abundance of 7.2%, and AlphaFold3 trimer compatibility); predicted disulphide cross-links were not demonstrated biochemically; and the interspace composition remains poorly characterised . Small ΔcsxC imaging cohorts (8 spores negative stain) limit quantitative strength, and stress testing beyond heat (65–95 °C) did not reveal fitness costs of losing the envelope, leaving its physiological role open—though the paper acknowledges the lab may not reproduce relevant stresses. BGPT infers that the convergent-evolution claim (Bacilli ExsY/CotY vs Clostridia CsxA/CsxC) is plausible but structurally inferential, resting largely on AlphaFold3 predictions without direct structures for the comparison proteins.
