The preprint reports that Poaceae (grasses) have undergone evolutionary rewiring of the anthocyanin biosynthesis pathway: (1) a Poaceae specific anthocyanin activator MYB lineage (ncaMYB), (2) an independent anthocyanin related GST lineage (BZ2) replacing the widespread TT19/An9 clade, (3) lineage-specific losses/gains in proanthocyanidin genes (ANR clades, LAR loss pattern), and (4) apparent wholesale loss of anthocyanin biosynthesis components (ncaMYB, BZ2, ANS) in Brachypodium β conclusions supported by phylogenomics, synteny, and expression/coexpression analyses using public genomes and RNA-seq [see primary source].
Key reproducible resources: GitHub scripts and pipelines used are publicly available (Poaceae_ABP repository) and RNAseq sources are from SRA (Additional File 1) enabling reproduction of the in silico analyses.
The paper presents a coherent and well-documented in silico case that anthocyanin biosynthesis regulation in Poaceae has been remodeled via independent lineage evolution (ncaMYB, BZ2) and lineage-specific structural gene losses (ANS, LAR patterns), with a provocative claim that Brachypodium has lost anthocyanin biosynthesis. The combination of phylogenetics, synteny, and coexpression provides substantial support; however, the major limitation is the absence of direct functional or biochemical validation. With targeted wet-lab confirmation (complementation, metabolomics, binding assays), the conclusions would become much stronger and mechanistically convincing.
Actionable reproducible resources: GitHub repo with scripts: https://github.com/ajones292/Poaceae_ABP (pipeline for gene detection, phylogeny, presence-absence matrices) β re-running the published pipelines on public SRA data should reproduce the main in silico findings
