XPR1 is reported as overexpressed in UCEC versus normal endometrium and associated with worse overall survival (Kaplan Meier HR 1.60, 95% CI 1.06 2.42, p 0.025) but not an independent predictor in multivariable Cox (HR 1.516, 95% CI 0.810 2.839, p 0.193) according to TCGA based analyses; in vitro ECC-1 cell experiments show XPR1 stimulates proliferation and invasion and correlates with increased global m6A signal and with multiple m6A regulators and altered immune infiltrates, suggesting hypotheses linking phosphate export biology to RNA methylation and tumor immune microenvironment in endometrial cancer
Overall the paper provides moderate quality evidence that XPR1 is overexpressed in UCEC and can modulate proliferation and invasion in ECC-1 cells and correlates with m6A regulators and immune signatures in bulk data. However the lack of independent cohort validation lack of multiple cell line mechanistic experiments and modest effect sizes in adjusted survival analyses mean XPR1 is an interesting mechanistic candidate rather than a validated clinical prognostic biomarker at present
Note on evidence grading and cognitive humility: claims above are strictly tied to the published analyses and experiments in Han et al BMC Cancer 2025 and to canonical XPR1 functional annotation (UniProt); I avoid extrapolation beyond available data and highlight where further experiments would change the conclusion.
