The introspected bias is real and material to the bronchiectasis experiment design I produced: I built a full protocol around mechanistic plausibility (CSF→NF-κB survival signaling ) and a strongly positive published cohort, without any visibility into unpublished null longitudinal CSF datasets. The positive cohort evidence is genuinely strong but carries risks my design only partially corrected: tertiary-center recruitment, 12-month follow-up, and culture-based infection definitions . Similar sensitivity/specificity asymmetries exist elsewhere in the supporting literature I lean on (e.g., upper-airway dysbiosis predicts severe exacerbations ~3-fold but lacks paired lower-airway sequencing and European-only generalizability ), meaning my evidence base is enriched for effects that survived publication.
Concrete corrections: (1) pre-register the sustained-CSF hypothesis with a formal falsification criterion so a null result cannot silently vanish; (2) treat moderate mechanistic cell-line signals (GM-CSF→IKKβ in HUVEC/TF-1 models) as hypothesis-generating only, not as design justification ; (3) note that even sensitivity of a bedside biomarker for chronic infection falls to ~72% at the informative cutoff — the signal-to-noise in my extrapolations is lower than the citation counts suggest.
