Grintsevich et al. combine pyrene-actin kinetics, co-sedimentation, subtilisin limited proteolysis, custom MetO44/Met44 antibodies, and two-colour TIRF microscopy to show that cofilin suppresses Mical's NADPH consumption and oxidation rate yet dramatically accelerates disassembly of Mical-oxidized filaments β a counterintuitive result they resolve by arguing that cofilin depletes F-actin (Mical's substrate) faster once oxidation has weakened inter-protomer contacts .
Key quantitative findings: purified Mical-oxidized actin has a critical concentration of 1.1 Β΅M Β± 0.25 (pH 7) versus ~0.1 Β΅M for unoxidized actin, a roughly tenfold destabilization . Even lightly oxidized copolymers (11% ox-actin) accelerated cofilin severing 22-fold (Student's t-test P = 0.004, n = 132 copolymers and 112 unoxidized filaments across 3 repeats), and cofilin binding was enhanced on disassembly-resistant crosslinked Q41C filaments (P = 0.0005) . TIRF showed fully oxidized filament stretches dismantled within the ~30 s solution exchange time when 10 nM cofilin was added, while buffer or either effector alone had weak effects.
In vivo, Drosophila bristle remodelling by Mical was suppressed by removing one copy of cofilin (twinstar) and enhanced by a hyperactive cofilin S3A transgene; effects depended on actin Met44 and were reversed by the reductase SelR . Transheterozygous cofilin/Mical mutants and gain/loss-of-cofilin modulation of PlexA-Mical axon guidance (n = 100 hemisegments or 480 longitudinals per genotype; chi-square and t-tests, P < 0.0001) place the pair in the same Semaphorin-Plexin pathway.
The finding explains why cofilin β a weak severer alone β becomes potent when actin is redox-modified. The paper's own statistics section candidly reports no pre-registered sample sizes, no randomization, and no blinding; effects were large and reproduced across two laboratories with independent reagents, which partially mitigates but does not eliminate observer-bias risk. Crosslinking experiments cleverly disentangled binding from disassembly, but the oxidized-actin mechanism (weakened inter-protomer contacts) remains inferred from fragmentation behaviour rather than directly visualized; later cryo-EM work on cofilactin supports the general concept of cofilin-driven conformational weakening . Blindspots: no mammalian in vivo validation, no quantification of physiological oxidation levels, and possible confounding from other actin-binding partners not tested.
