     Quick Explanation

SYDE1 and SYDE2 are established here as JNK-selectively phosphorylated RhoGAPs with in vitro GAP activity toward RhoA, Rac1, and Cdc42, and with GAP-partially-dependent effects on HEK293 spreading, focal adhesions, and directional migrationβ€”yet no bulk GTP-loading changes in cells were detected, leaving their authentic cellular targets unresolved

     Long Explanation

    What the paper shows

Song, An, Lou, and Turk (Yale) resolve conflicting screens on two orphan RhoGAPs. In vitro, purified human SYDE1 (172–587) and mouse SYDE2 (507–944) stimulate GTP hydrolysis by RhoA, Rac1, and Cdc42; Arg-finger mutants (R369A/R707A) reduce activity, confirming catalytic dependence . Kinase assays with activity-normalized MAPKs show SYDE1 Ser578 phosphorylated solely by JNK and SYDE2 Ser935 preferentially by JNK1 (weakly also by ERK5/ERK2) . Full-length phosphorylated SYDE2 showed no change in catalytic GAP activity, and S578E phosphomimetic SYDE1 only slightly increased RhoA-directed activityβ€”so JNK phosphorylation likely regulates SYDE proteins indirectly (localization/interactions), not catalysis itself.

    The surprising negative result

Despite clear in vitro activity, G-LISA assays in a clonal SYDE1 KO/rescue Swan 71 trophoblast system (n=5) and HEK293 overexpressors (n=3) found no bulk GTP-loading change for RhoA, Rac1, or Cdc42 . The authors honestly flag this as the paper's central unresolved question. Computational work had predicted RhoA (and possibly RhoB) docking to SYDE RhoGAP domains , yet proximity-labeling screens previously implicated Cdc42/RhoQ/RhoJβ€”consistent with the in-vitro-promiscuity caveat the authors themselves raise.

    Cellular phenotypes: real but overexpression-dependent

Stable GFP-tagged SYDE1/2 in HEK293 increased cell area, decreased circularity, raised focal-adhesion number, and made migration more persistent (lower mean directional change rate, higher linearity); SYDE1's effects on spreading and adhesion were lost in the R369A mutant, whereas SYDE2 RA only trended toward weaker phenotypes . Blind-spots the paper acknowledges: no endogenous SYDE2 perturbation, no primary cells or in vivo work, no direct test of JNK phosphorylation on cellular phenotypes, and the truncated SYDE1 construct excludes autoinhibitory N/C-terminal regions proposed by prior mouse SYDE1A work. What would change the conclusions: FRET-biosensor or biosensor-based spatial GTPase measurements showing localized RhoA/Rac1/Cdc42 attenuation by SYDE1/2; if FRET also shows nothing while RA mutants still phenocopy, non-catalytic scaffold functions would become the default explanation.

     BGPT Paper Review

    Study Novelty

    60%

First panel-level in vitro GAP activity measurement and JNK-selective phosphorylation mapping for SYDE1/2, but prior screens already implicated these GTPases and phenotypes; novelty lies in consolidation, not discovery.

    Scientific Quality

    70%

Rigorous activity-normalized kinase assays, catalytic-mutant controls, and honest reporting of null G-LISA results strengthen quality; weaknesses are overexpression-only cellular conclusions, phosphomimetic limitations, and truncated SYDE1 construct.

    Study Generality

    50%

Findings are solid but confined to two proteins, two immortalized cell lines, and in vitro assays; no generalized mechanistic principle beyond the GAP-promiscuity caveat, which applies broadly.

    Study Usefulness

    60%

Establishes a firm biochemical baseline (substrates, phosphorylation sites, catalytic residues) and reusable KO/rescue and imaging workflows that directly enable future target-identification studies.

    Study Reproducibility

    60%

Detailed methods (constructs, buffers, imaging settings, statistical tests) support replication; but no deposited raw data, image-analysis scripts, or code links limit full reproducibility.

    Explanatory Depth

    50%

Biochemical mechanism is well supported, yet the causal chain from GAP activity to morphology/migration remains correlative and partly GAP-independent; authentic cellular targets unidentified.

     Hypothesis Graveyard

'SYDE1/2 reduce bulk RhoA/Rac1/Cdc42 GTP-loading in cells' β€” directly falsified here by null G-LISA in KO/rescue and overexpression systems.

'Worm Syd-1 catalytic mechanism predicts mammalian SYDE GAP substrate' β€” undermined because worm Syd-1 lacks the conserved Arg finger and is catalytically inactive.
