     Quick Explanation

Millet et al. show C. elegans prefers easier-to-eat normal bacteria over elongated ones at equal density, that higher density of effortful food reverses preference, and that indifference points follow a 2/3 discount factor as predicted by standard effort-discounting modelsβ€”extending discounting-like behavior to a lower invertebrate .

     Long Explanation

    Core finding and evidence

This eLife study (published December 2025) demonstrates that C. elegans exhibits effort-discounting-like food choice. The authors elongated Comamonas bacteria with cephalexin, showing that: (1) pumping frequency is unchanged (p=0.92, d=0.03, n=66/60), but fat stores drop (p=3.2e-04, r=0.43), indicating more pumps per unit nutrient; (2) nutrition (protein/carb/lipid, all p>0.9) and odor attractiveness (azide-trapped T-maze, p=0.36, n=108) are unaffected; (3) in free-choice T-mazes, worms prefer normal bacteria at equal OD (I=+0.10, p=1.69e-03, n=137), are indifferent at OD 1.5 elongated (I=0.03, p=0.28, n=106), and prefer elongated at OD 2.0 (I=-0.17, p=2.06e-07, n=73) .

    Strengths: quantitative economic prediction

The standout contribution is the derivation of a discount factor d(E)=2/3 from the indifference point, then out-of-sample prediction of novel indifference points at OD ratios 2:3 and 0.5:0.75 β€” both confirmed (one-way ANOVA across three novel ratios: F=0.83, p=0.44, Ξ·Β²β‰ˆ0) . Patch-leaving kinetics show leaving rates (k_FB, k_BO) increase on elongated food while entry rates are unchanged (MANOVA p=8.77e-11), supporting a local-assessment mechanism rather than cross-arm comparison .

    Critical caveats

Dopamine paradox: dop-3 mutants show indifference (p=0.019), but cat-2 and dat-1 mutants are normal β€” authors acknowledge this is puzzling and consistent only with dop-3-specific signaling. Confounds: cephalexin elongation is a single manipulation; subtle unmeasured bacterial differences cannot be fully excluded. N2 laboratory bias: N2 has 50+ years of lab adaptation to easy-to-eat E. coli; only 5 wild isolates were tested with high variance, and DL238 behaved like N2 while JU775 was indifferent β€” genetic background effects are unresolved . Interpretive gap: "effort discounting-like" is phenomenological β€” whether the 302-neuron system computes subjective value or simply follows a local stimulus-response rule remains untested. This study builds on prior utility-maximization work showing GARP-consistent choice . COI: senior author has a financial interest in InVivo Biosystems (ScreenChip used in Fig. 1A) β€” low impact on the central T-maze findings but disclosed.

    Verdict

Strong, well-controlled, quantitatively predictive behavioral study. The mechanism-level claims (dopamine role, subjective value) remain correlational and warrant caution; the behavioral phenomenon itself is robust.

     BGPT Paper Review

    Study Novelty

    80%

First demonstration of effort-discounting-like behavior in a lower invertebrate with quantitative model-based prediction of novel indifference points; extends discounting phylogeny but builds on prior GARP work in the same system.

    Scientific Quality

    80%

Exhaustive controls (nutrients, odor, pumping rate), full statistical reporting with effect sizes, out-of-sample predictions confirmed. Weaknesses: dop-3/cat-2/dat-1 inconsistency unexplained, only 5 wild isolates with high variance, COI in pumping-assay hardware.

    Study Generality

    80%

Extends cost-benefit decision-making frameworks across ~600 My of evolutionary distance; implications for neuroeconomics, behavioral ecology, and discounting models broadly, though ecological validity in natural habitats is untested.

    Study Usefulness

    80%

Provides a genetically tractable system with a quantified discount factor for dissecting effort-based choice; immediately useful for C. elegans neurogenetics and comparative discounting research.

    Study Reproducibility

    80%

All raw data in Source data 1, analysis code in Source code 1, detailed methods, CGC strains, and full statistical table (Table 1) support independent replication.

    Explanatory Depth

    70%

Strong behavioral-economic formalization and kinetic mechanism, but neural implementation (how 302 neurons compute discounted value, dop-3's specific role) remains speculative.

     Hypothesis Graveyard

Effort discounting requires a centralized brain or large neuron count: falsified by this study β€” 302 neurons suffice for discounting-consistent choice.

Dopamine uniformly increases effort tolerance across animals (rodent model): weakened β€” in worms, dop-3 loss increases effort tolerance, opposite to rodent pharmacological effects, suggesting chronic-vs-acute or receptor-subtype differences.
