This study achieved the first purification to homogeneity of E. coli glucosamine synthetase (L-glutamine:D-fructose-6-phosphate amidotransferase, the glmS gene product), a 70.8 kDa subunit enzyme whose lability had defeated earlier attempts. A 400-fold purification from 500 g of cells yielded ~10 mg of >90% pure enzyme at 7.63 units/mg β 30-fold above the original Gosh preparation and 11-fold above the best prior bacterial preparation .
Key mechanistic finding: labeling with the glutamine analogue [6-14C]DON, followed by CNBr cleavage and Edman degradation, placed essentially all radioactivity at cycle 1 β identifying the NH2-terminal cysteine as the glutamine-binding, Ξ³-glutamyl thioester-forming residue. Notably, the gene-predicted initial methionine is removed in vivo .
The enzyme showed no NH3-dependent activity and no detectable glutaminase or reverse reaction β supporting tightly coupled amidotransfer rather than free-ammonia chemistry. DON stoichiometry was Fru-6-P dependent (0.47 vs 0.87 mol/subunit), suggesting half-of-the-sites reactivity and Fru-6-P-induced conformational changes; the authors themselves flag the unexplained IAAm asymmetry as an open question . Blindspots include oligomeric state ambiguity: gel filtration gave 170β193 kDa, sedimentation-derived calculations gave 127 kDa, and cross-linking produced a 210 kDa species with no detectable dimer band β internally inconsistent data the dimer hypothesis only partially resolves. Structural evidence was chemical (alkylation, sequencing) rather than direct 3D visualization.
This work became the enzymological anchor for the GlmS/N-acetylglucosamine pathway. Its DON-labeling logic prefigured later work on mechanism-based glutamine-analogue inhibitors; the Candida albicans study by the same school (FMDP, Ki 1.0 Γ 10-7 M) built directly on the active-site cysteine concept . Modern coupled assays for GlmS inhibitor screening still descend from this characterization . What would change the conclusions: a crystal structure contradicting the N-terminal cysteine placement, or demonstration that NH3 sustains activity under Fru-6-P-bound conditions.
