The paper challenges a binary on/off model of AIF activation. In MEFs, the dimerization-defective hAIF 3M variant (E413A/R422A/R430A) impaired galactose growth (P=0.0415 vs WT), reduced supercomplex incorporation of complexes I and III to ~90% and ~70% of WT, and raised normalized mitochondrial ROS by ~60% β yet preserved near-WT respiration, whereas AIF KO caused severe respiratory collapse . This graded phenotype is the key conceptual advance beyond earlier work establishing AIF-CHCHD4 as essential for respiratory-chain biogenesis .
The most compelling mechanistic result is reciprocal rescue: CHCHD4 increased 3M catalytic efficiency ~38-fold to near-WT levels, and the N-terminal CHCHD4 peptide accelerated hydride transfer ~400β450-fold in WT and 3M variants, without altering redox potential (Em β -358 mV) . Translationally, the N-CHCHD4 peptide restored pathogenic R422QΞ77 CTC lifetime ~16-fold and lowered Kd ~5-fold despite minimal conformational stabilization . This aligns with recent cryo-EM evidence that CHCHD4 engages the reduced AIF dimer C-terminal Ξ²-sheet platform .
Everything is in immortalized MEFs and purified Ξ77 truncants β not neurons, the clinically relevant context for AIFM1 disease; the peptide rescue used 1:3 molar excess in vitro with no cellular delivery shown; oxidized 3M ITC was unmeasurable due to instability; and no DRS substrate import flux was directly measured. Critically, the paper offers no in vivo or patient-cell validation of peptide rescue. Falsification would require showing that a dimerization-defective AIF fully restores supercomplex assembly and normalized ROS, which would collapse the graded-plasticity model . Confidence in the mechanistic claims is moderate-to-high (quantitative, multi-method, n=3+ replicates); confidence in therapeutic relevance is low.
