The strongest dataset attributable to Katherine Wang is a cross-species mechanistic study of human developmental tempo. Its scientific architecture is unusually complete for a single first-author paper: discovery (species comparison), causal testing in both directions, mechanism rescue, and generalization across germ layers.
Discovery: Comparative profiling of mouse vs human stem cell-derived presomitic mesoderm showed mouse cells maintain a higher cytosolic NAD+/NADH ratio (SoNar imaging, Fig. 1B) and higher [4-2H]glucose M+1 labeling into glycerol-3-phosphate (Fig. 2F-H), pinpointing low GPD1L as the shuttle bottleneck .
Bidirectional causality: Raising NAD+/NADH with LbNOX shortened the segmentation clock period by ~30 minutes; lowering it with EcSTH lengthened the period by ~20 minutes (fig. S3I-K). A dose-like bidirectional effect is a hallmark of genuine mechanism rather than an artifact of a single perturbation .
Target rescue and generalization: CRISPRa activation of GPD1 or GPD1L raised G3P M+1 flux, shortened the clock period by ~40β50 minutes, and increased basal/maximal respiration including complex I-independent respiration (Figs. 3B-D, 4A-E) . The effect extended beyond PSM: SOX17-tdTomato endoderm reporter activation appeared one day earlier and PAX6::GFP neural progenitor activation ~two days earlier, both P < 0.0001 by one-way ANOVA with Tukey correction (n = 3 replicates; Fig. 5G-J) .
Critical qualification: Notably, the author reported that GPD1/1L overexpression did not change MSGN1-Venus induction timing despite faster clocks and cell cycles (E5, mixed status) β an honest internal boundary on the mechanism, indicating redox tempo and fate-acquisition timing can be uncoupled .
What would change this assessment: no independent replication of the human embryo in vivo inference (PCW3 clusters) is included, sample sizes for most experiments were not reported in the extractable record, and metabolic tracing relies on a single labeling substrate ([4-2H]glucose). Confirmation with orthogonal NAD+ flux assays or human embryo primary data would raise confidence; failure of GPD1L knockdown to slow the human clock would weaken the central claim.
