Adam Lange is a senior group leader (Leibniz-FMP Berlin; Charité environment) specializing in solid-state NMR (ssNMR) of insoluble supramolecular assemblies and membrane proteins, frequently combined with cryo-EM and MD. His citation profile — h-index 41, 6,334 citations, 142 papers — places him among the established leaders in biomolecular ssNMR. He held a DFG Emmy Noether Fellowship (2012) and has since led major structural-mechanics projects on Type III secretion needles, bacteriophage SPP1 tails, amyloid/prion fibrils, potassium channels, and VDAC, many in the se structural biology world of the Max Planck/Berlin tradition.
Interpretive caution: citation metrics favor long-career NMR methodologists (his technique-development papers accrue citations over decades); they do not guarantee that any single recent preprint is correct.
Lange's 2025–2026 work exemplifies a mature integrative strategy. In the MthK potassium channel study, cryo-EM (106,517 initial particles; 3.2 Å C4-symmetric map), ssNMR (1H-detected at 55 kHz MAS with 15NH4+ ion surrogates), and 3×500 ns MD together show charybdotoxin anchoring K27 into the S0 site while the selectivity filter stays non-collapsed, with S2/S4 ion occupancy confirmed by ammonium NMR; all maps/assignments deposited (EMD-56657, PDB 28NO, BMRB 53511). Blindspots reported by extraction: reliance on AlphaFold3 templates for classification, ambiguous S3 occupancy, and pore-domain truncation in ssNMR.
In the EcMscL mechanosensitive channel work, cryo-EM (3.5–3.7 Å) and ssNMR in liposomes converge on a closed state for WT and G22S, but ssNMR assignment coverage was only 43% (WT) and ~20% (G22S), and no open-state structure was captured — the open-transition mechanism remains inferred from dynamics, not observed.
Lange's 2012 sparse-[1/2-13C]-glucose labeling method achieved ~80% bi-directional sequential assignment and 73 long-range restraints for the Salmonella T3SS needle — a generalizable ssNMR advance validated against PDB 2LPZ. His FCHO1/FCHO2 IDR work shows he extends beyond method into evolutionary cell biology, with transferable-helix causality established by chimera and mutation experiments. Note: the amphipod, hydrology, FTLD, HIV, and soybean-segmentation records matched to this author name are almost certainly name-collision artifacts from author-disambiguation, and were excluded from scoring.
Lange is a highly rigorous, method-driven structural biologist whose integrative designs, rigorous validation (MolProbity, deposition discipline), and honest limitation statements are hallmarks of strong science. Known unknowns: open-state channel structures remain unobserved; AlphaFold3 dependence introduces possible circularity; and in-vivo validation of in-vitro assembly claims (FCHO) is not yet endogenous. If independent labs fail to reproduce his deposited structures — or if AlphaFold3-biased classification is shown to have forced wrong toxin orientations — the assessment would downgrade.
