The authors report a practical workflow that converts otherwise βnon-assemblingβ HIV-2 CA into pelleting, EM-observable capsid-like assemblies by switching the inducer from NaCl to potassium glutamate (KGlu), with IP6 enhancing efficiency and pH shifting morphology (more curved/smaller at lower pH; more tubular/extended at higher pH).
They then test interface logic by mutating divergent HIV-2 residues to HIV-1 equivalents (and capsomere-destabilizing βone-formβ disruption designs). Assembly kinetics and morphology shift as predicted for capsomere-specific stability effects, although multiple phenotypes still show residual pleomorphism and occasional amorphous aggregation.
The work most immediately improves experimental accessibility to HIV-2 CA assembliesβenabling systematic biochemical testing of mutants and interface logic under a defined buffer regimeβwhile also suggesting that βanion identity + pHβ may be first-order control parameters for pleomorphism.
