Scope and verdict. This is a well-constructed expert review (not primary research) that reframed filamentous cyanobacteria as a premier model of bacterial multicellularity, built on three pillars: cell-cell adhesion via a continuous filament-wide outer membrane, intercellular communication through septal junctions, and terminal differentiation of photosynthetic vegetative cells into N2-fixing heterocysts .
Key content strengths. The review correctly emphasized a striking ultrastructural fact: the outer membrane of heterocyst-forming cyanobacteria is continuous along the entire filament and does not invaginate at septa, even though cytoplasmic membranes and peptidoglycan layers do . It also flagged a genuine puzzle: Anabaena sp. PCC 7120 lacks obvious orthologues of canonical proteobacterial porins, and its LPS is atypical (lacking phosphate, KDO, and heptose), leaving outer-membrane permeability under-characterized .
How the review's framework has held up. Two supplied papers extend it. FraI in Nostoc punctiforme is now shown to be required for nanopore-dependent communication: a ΞfraI mutant shows reduced motility, impaired cell-cell exchange, and failure to differentiate heterocysts while still forming akinetes . The predicted developmental gene-regulation layer was confirmed by a 2026 mBio study: NfsS phosphorylates NfsR (Ser51), NfsS binds sigma factors SigC/SigE, ΞnfsR abolishes heterocysts and diazotrophic growth, and 276 genes are differentially expressed, including nifH, nifB, and fdxH .
Critical limitations of the review. (1) It is narrative and citation-dense but largely pre-genomic in mechanistic depth; the cognate phosphatase for NfsR and the upstream nitrogen sensor remained undefined even 17 years later . (2) Mechanistic claims about Anabaena porins rested on absence of orthologues, an argument from silence. (3) Single-model bias: most conclusions derive from two strains (Anabaena sp. PCC 7120, N. punctiforme PCC 73102), limiting phylum-wide generality. (4) The correlational enrichment of partner-switching systems in filamentous genomes (2.4-9.4-fold vs unicellular) does not establish causation of multicellularity .
What would change the conclusions. Demonstration that filament integrity persists without a continuous outer membrane, that heterocysts form without NfsR/SigC/SigE signaling under defined conditions, or that nanopore-independent metabolite exchange suffices for diazotrophic growth would each falsify a core pillar.
Note: bar heights for filamentous classes reflect only the reported 2.4- to 9.4-fold range endpoints plotted as an illustrative scale; the exact per-morphotype means were not supplied.
