The study extends the authors' earlier finding that DNA damage activates CUL4B to target HUWE1 for degradation () by identifying the substrate receptor: RBBP7. Evidence chain: (1) RBBP7 co-immunoprecipitates with CUL4B, HUWE1, DDB1, and Cul4A/B in HeLa S3 stable lines; (2) GST-RBBP7 pulls down HUWE1 and DDB1 directly; (3) RBBP7 knockdown raises HUWE1 levels and extends HUWE1 half-life in CHX chase, while overexpression shortens it; (4) recombinant Flag-HA-CUL4B complex plus GST-RBBP7 ubiquitinates GST-HUWE1(1-2500) in a dose-dependent, ATP/E1/E2-dependent in vitro reaction; (5) downstream substrates respond as predictedβRBBP7 depletion lowers BRCA1/BARD1 (via HUWE1 upregulation) and lowers MCL-1, rescued by MG132 or by co-depleting HUWE1.
Strengths: reciprocal IP, endogenous co-IP, GST pull-down, and in vitro reconstitution together form a coherent interaction-to-function argument; two independent siRNAs were used for RBBP7; MG132 epistasis correctly places MCL-1 loss downstream of the proteasome. The structural logic fits CRL4 biology, where WD40 DCAFs recruit substrates ().
Weaknesses and blind spots: (i) The key substrate-recognition step is inferred, not provenβno RBBP7 mutant lacking HUWE1-binding, no domain mapping, and no HUWE1 ubiquitination-site identification beyond βsomewhere in residues 1-2500.β (ii) In vitro ubiquitination used a truncated GST-HUWE1 fragment from baculovirus; the later full-length HUWE1 cryo-EM structure (ring-shaped alpha-solenoid, 3.1 Γ ) shows N-terminal architecture is central to ligase loading, so truncation effects cannot be excluded (). (iii) No endogenous HUWE1 ubiquitination assay, no DNA-damage time-course with RBBP7 depletion, and no in vivo or patient dataβnotable given HUWE1 dosage sensitivity causes X-linked intellectual disability with tissue-specific XCI effects in humans (). (iv) RBBP7 is also a core histone chaperone in NuRD/Sin3/PRC2 complexes; disruption could affect BRCA1/MCL-1 indirectly via transcription, a confound the paper acknowledges but does not test. (v) Only cancer-cell lines (HeLa, HEK293T); the therapeutic framing (βtargeting CRL4B-RBBP7β) is speculative, and later work shows HUWE1 loss can be tumor-promoting in APC-mutant colorectal cancer (), making unidirectional βHUWE1-down = pro-survivalβ framing oversimplified. RBBP7 depletion promoting apoptosis was explicitly left unverified by the authors.
What would change the conclusion: a binding-defective RBBP7 mutant failing to degrade HUWE1, endogenous ubiquitination of full-length HUWE1 with mapped lysines, and rescue in DNA-damaged primary cells. If RBBP7 knockdown stabilizes HUWE1 equally in CUL4B-null cells, the model would be falsified.
