The authors present evidence that TGF-Ξ² locally imprinted by physical neuron macrophage contact drives a nerve associated macrophage phenotype (sNAM) required for nerve regeneration, using complementary in vitro cocultures, fateβmapping and conditional Tgfbr2 deletion in CX3CR1+ macrophages
The paper presents strong, multi-angle evidence that locally activated TGF-Ξ² is a major instructive cue for the sNAM transcriptional and phenotypic program and that this imprinting matters for cutaneous nerve regeneration in the ear punch model. The design combines perturbations at ligand activation (integrin blockade), receptor signaling (SB-525334, Tgfbr2 flox), transcriptional readouts (bulk RNAseq) and functional outcome (reinnervation), which constitutes a robust demonstration of causality in the studied system
However, the conclusion that sensory neurons are the principal in vivo source of the bioactive TGF-Ξ² that imprints macrophages is plausible but not unequivocally proven. The authors explicitly note alternative activation paths (autocrine macrophage TGF-Ξ² activated by neuron metalloproteases or Schwann cell integrins) and present transcriptomic evidence for integrins and MMPs in macrophages but lack, for example, conditional neuron-specific Tgfb1 depletion or in situ proximity-based activity sensors that would definitively attribute activation to neurons alone
Spatial map showing pSMAD2/3 intensity overlaid on whole mount dermis images with macrophage and neuron markers (Ξ²3 tubulin and F4/80/CX3CR1) in control versus Tgfbr2 conditional knockout: would directly show the spatial confinement of active TGF-Ξ² signaling to nerve associated macrophages.
This is a high-quality, mechanistically focused study that convincingly identifies local TGF-Ξ² activation during neuron macrophage contact as a key instructive cue for formation and maintenance of nerve-associated macrophages and links this imprinting to functional nerve regeneration in skin; further mechanistic work to pin the exact cellular source(s) and full data release will strengthen translational prospects.
