Cholesterol: In Huh7 cells, acute free-cholesterol accumulation enlarged and promoted fusion of lipid droplets, induced PLIN2 degradation, and affected ApoB association with droplets in a Rab18-dependent manner. This is an ERβlipid-droplet trafficking phenotype, not evidence for late-endosome escape of siRNA.
Rab18 and siRNA: Rab18 siRNA was used experimentally to deplete Rab18. Depletion removed Rab18 from ER tubules and expanded ER sheets, with reported CLIMP-63 area increasing from approximately 30% to 60β80% of cell area; rescue with GFP-Rab18 supported specificity. These results concern Rab18 function and ER architecture, not siRNA escape from endosomes.
What is missing: No supplied study measures late-endosomal cholesterol, Rab18-dependent endosomal membrane permeabilization, cytosolic siRNA delivery, endosomal escape efficiency, or a causal cholesterol-manipulation/escape relationship. Therefore, the scientifically supported conclusion is: cholesterol can modulate Rab18-linked ERβlipid-droplet behavior, but a Rab18βcholesterol mechanism for siRNA escape is presently un demonstrated by these records.
Important distinction: βsiRNA knockdown of Rab18β means siRNA is being used to reduce Rab18 expression; it does not mean that Rab18 mediates the escape of siRNA from late endosomes. Direct testing would require separating uptake from cytosolic release and measuring functional knockdown alongside endosomal localization and cholesterol perturbation.
