Luff et al. identified TACI as the only specific co-purifying interactor of tagged BAFFR (found only in BAFF-stimulated cells), then re-examined TACI's role. Prior literature concluded TACI negatively regulates B cell survival because Tnfrsf13b-/- mice have expanded B cells . The authors elegantly resolve the confound: TACI-KO mice have ~3-fold higher serum BAFF, but in 50:50 mixed bone marrow chimeras serum BAFF is normalized, hyperplasia disappears, and yet TACI-deficient (CD45.2+) MZ B cells are selectively reduced in proportion and number while FO and T2 B cells are unaffected β establishing a cell-intrinsic developmental defect, not a survival or homing problem (EdU pulse-chase over 42 days and 1-hour adoptive transfer showed TACI-KO MZ cells actually homed more efficiently).
Functionally, B6:129 chimeras immunized with TNP-Ficoll showed greatly reduced TACI-deficient IgMb and IgG1b responses while the internal WT 129 allotype controls were unaffected, plus reduced TACI-deficient plasma cell expansion .
RNA-seq is strikingly specific: 686 DEGs in TACI-deficient MZ B cells vs only 34 in FO B cells, with loss of MZ signature (NES β3.009) and gain of FO signature (NES 3.008), both padj = 1.16e-47 . FOXO1-activated targets were upregulated (NES 1.867), consistent with reduced pAKT-T308/S473 and reduced pS6 measured by flow cytometry. TACI constitutively associated with PI3KΞ΄ (p85Ξ±/p110Ξ΄) and mTOR, ligand-independently. This aligns with the established AKT-FOXO1 axis in MZ development .
Critical caveats: the PI3K-AKT-FOXO1 mechanism is inferred from correlative phospho-flow and GSEA; no rescue experiment (e.g., forced AKT activation or FOXO1 haploinsufficiency in TACI-KO MZ cells) directly proves causality. The ligand driving TACI signaling (BAFF vs APRIL vs syndecan-2) remains unresolved β the authors themselves note APRIL-KO mice have normal MZ numbers. Human relevance is untested despite CVID-linked TNFRSF13B mutations . Data deposition (GSE300247, PXD065033) supports reproducibility.
