Using baculovirus expression in SF9 cells and single-particle cryo-EM, the authors (using the endonuclease-dead D97A mutant to prevent RNA degradation) determined five structures: RNA-free core (2.78 Γ , PDB 8P1J), core+endonuclease (3.24 Γ , 8P1K), full-length (3.32 Γ , 8P1K/EMD-17353), 5β² vRNA-bound core (2.79 Γ , 8P1M), and a 5β² vRNA+NTP-bound state (3.23 Γ , 8P1N) . The full-length structure resolves the previously invisible cap-binding domain (residues 1704β1825, with putative cap-sandwiching residues Y1710/Y1725 ~8 Γ apart), a bipartite midlink domain, and a C-terminal helical domain (1921β2151) with no Foldseek homologs β a conformation the authors state is incompatible with transcription and novel among cap-snatching polymerases .
The most mechanistically interesting result is motif E (1166β1186, consensus Ex2S): it adopts four distinct conformations (Ξ²-sheet, two helical states, ordered loop) across the structures. NTP binding reorients E1177 to coordinate an active-site MgΒ²βΊ non-canonically alongside D1098/1099, D972 β interpreted as a pre-replication initiation state and a decision point between transcription and replication .
Strengths: high-resolution maps, thorough classification workflow (cryoSPARC v4 + Relion 3D classification without alignment) to recover rare full-length particles, and complete data deposition. Limitations: this is an unreviewed preprint with visible OCR-quality text artifacts; the full-length structure contained no RNA despite being prepared with promoters, capped RNA, and NTPs, so its physiological relevance to an actual transcription/replication intermediate is inferred, not demonstrated. The NTP identity is ambiguous (weak ribose/base density; authors acknowledge a possible mixture), and all structural states were trapped via PEGylation, which could bias conformer populations. Functionally, the work is purely structural β no polymerase activity assays accompany the structures; concurrent work (ref. 23, Durieux Trouilleton et al.) independently reported overlapping HTNV structures, reducing exclusivity of the findings. Follow-up biochemical work confirmed a conserved terminal uridylyl transferase activity of HTNV L that must be controlled for in interpreting such polymerase complexes . What would change the conclusions: a full-length structure with ordered promoter RNA and a biochemically validated replication state, and replication of motif E rearrangements in a homologous system.
