Zhang et al. (PNAS, 2012) identify TCTP (translationally controlled tumor protein) as a participant in the DNA damage response (DDR) using normal human fibroblasts (AG1522), ATM-mutant lines (AT5B1, AG4405), U2OS and DNA-PKβmutant glioblastoma cells, plus C3H/HeJ mouse tissues
Low-dose 137Cs Ξ³ rays (10 cGy, 0.2 cGy/h) reduced micronuclei below spontaneous levels and up-regulated TCTP ~2-fold; acute 10 cGy gave ~6-fold nuclear enrichment by 4 h and 4β11-fold nuclear increases across 10β400 cGy . TCTP knockdown abolished repair kinetics: after 50 cGy, Ξ³H2A.X foci fell to 2.6 Β± 0.4 (3 h) and 0.7 Β± 0.3 (6 h) in controls but persisted at 9.6 Β± 1.0 and 8.6 Β± 0.4 in siTCTP cells (P < 0.001); Ku DNA-binding activity fell >50% (P < 0.001), and the radiation-induced G1 delay shortened from 14.5 h to ~9 h with attenuated p21 induction .
Strengths: convergent approaches (iTRAQ proteomics, siRNA, pharmacological inhibitors Ku55933/NU7441/PJ34, co-IP/MS, foci colocalization, clonogenic survival), multiple cell strains and 3D culture, in vivo mouse confirmation, and appropriate statistical rigor (P values, repeated experiments 2β6Γ) .
Limitations and unknowns: (1) The authors themselves note the TCTPβATM interaction may be indirect; targeted MS failed to detect TCTP phosphorylation at putative PI3KK sites (T39, S46, S53), so the upstream mechanism is unresolved . (2) The putative chaperone role for Ku translocation is inferred from interaction with Hsp60/70/90, not directly tested. (3) No quantification of mRNA vs. protein contribution to TCTP up-regulation. (4) Whether TCTP modulation affects the adaptive response (reduced MN below spontaneous rate) causally was not shown β knockdown experiments tested repair, not the adaptive-response phenotype itself. (5) Only two primary fibroblast strains plus tumor lines; cell-type generality beyond mice untested.
What would change the conclusion: demonstration that TCTP's effects on Ku are secondary to ATM/DNA-PK effects, a lack of colocalization at single-break resolution, or rescue experiments showing Ku overexpression restores repair in TCTP-depleted cells would reposition TCTP as a modifier rather than an essential DDR component.
