Guri et al. leverage 1,794 water samples (5,394 qPCR reactions) assayed for Pacific hake and 554 metabarcoded samples from the 2019 Joint U.S.–Canada hake acoustic-trawl survey, filtering to 371 samples where hake DNA was detected in both streams (Source). Eight mock communities of 39 fish species calibrate species-specific amplification efficiencies (Source), and a joint Bayesian model (Stan) expands hake qPCR concentration to the other 12 species using metabarcoding proportions (Source). Spatial smoothing via sdmTMB produces 3D maps across 0–500 m depth and 10° latitude.
eDNA depth patterns match known ecology: widow rockfish confined to ~50 m, sardine/anchovy at surface, northern lampfish spanning depths consistent with diel vertical migration, Dover sole near bottom (Source). Anchovy peaked at 43.5°N vs. sardine at 40–45°N, matching thermal niche differences. Species correlations cluster by ecological group (surface species Pearson ρ>0.50; midwater/deepwater all |ρ|<0.40). Richness and total eDNA peak near the Columbia River plume and Heceta Bank—known productivity hotspots (Source).
Note: The depth-profile figure is a schematic illustration of the qualitative patterns the authors describe, not a reproduction of their quantitative data (per-sample concentrations are in supplementary materials).
The authors acknowledge that no independent biomass cross-validation was performed—validation relies on agreement with literature distributions, which risks confirmation bias if literature ranges are coarse. Vertical transport (fecal deposition, carcass sinking) cannot be distinguished from true presence at depth, exemplified by sardine eDNA detected below 150 m in only 17/120 samples with 15 below 100 copies/L (Source). Dependence on a single reference species (hake present in 86.7% qPCR and 67.7% metabarcoding samples) limits extension to communities lacking a ubiquitous co-detected species. The GitHub link for the QM package is anonymized (www.github.com/anonymous), reducing reproducibility.
