The paper reports the first high-resolution structures of adenovirus capsids stalled before (E0, HAdV-C2 ts1 light, 3.3 Γ ) and during (E1, HAdV-C5 FC31 light, 3.0 Γ ) protease maturation, allowing a four-state comparison with genome-containing F0 and F1 maps .
Note: bar values are illustrative of the qualitative traceability described; the paper's quantitative anchors are the ANOVA p-values and penton-base grey value of 0.2 in E0 vs ~0.7 elsewhere.
Strengths: rigorous statistical treatment of map densities, gold-standard FSC resolution, public deposition (9TWZ/9TVV), and the elegant four-state design isolating genome-vs-protease effects. Authors acknowledge the HAdV-B7 VLP discrepancy (disordered IIIa/VIII there) may be species-specific or an artifact of recombinant production .
Blind spots: the L1-disorder mechanism is inferred from correlation, not mutagenesis; particle counts are not reported for statistical comparisons; no direct infectivity or mechanical-stability assay validates the metastability claim; and E0 derives from HAdV-C2 while E1 derives from HAdV-C5, introducing a serotype confound. A falsification test would be an E0-like capsid where L1 is cleaved but IIIa/VIII remain uncleaved β if vertex N-termini stay disordered, the L1 mechanism collapses .
A technically strong structural study with genuine mechanistic novelty β the IIIa 478-486 bridge and the VIII peptide-swap scaffold exchange are new observations. Interpretations about assembly error-correction and metastability priming are plausible but rest on correlational evidence and await mutagenesis and biophysical stability testing.
