The paper's strongest quantitative result is direct: switching the plasmid backbone from low-copy p15A to high-copy ColE1 increased constitutive circuit GFP/OD by approximately 20-fold, whereas the negative autoregulatory (NAR) TetR-feedback circuit changed only approximately 1.5-fold across all tested aTc concentrations (0β50 ng/mL) . Under arabinose-induced mCherry ribosome sequestration, increasing arabinose raised mCherry/OD and lowered GFP/OD, and sequester-NAR circuits maintained higher GFP/OD than sequester-constitutive at comparable loads .
These observations are framed analytically: contraction theory bounds trajectory deviation under ribosome perturbation by ¡δ/Ξ», so increasing the contraction rate Ξ» tightens robustness. The authors derive a ceiling Ξ» β€ Ξ³ (protein degradation rate) for constitutive expression and show NAR feedback adds a stabilizing term, exceeding that ceiling β at the cost of lower steady-state expression . Simulations with 100 uniform Β±30% ribosome perturbations showed smaller mean deviation and narrower spread for NAR .
Several gaps temper the conclusions. First, the copy-number experiment does not isolate resource availability: ColE1 also raises gene dosage and transcription, so the 20-fold constitutive change conflates template abundance with ribosome demand β a confound the authors acknowledge only implicitly. Second, the analytical bound ¡δ/Ξ» is never quantitatively compared to measured expression magnitudes; agreement with experiments is described as "phenomenological," so the theory is directionally, not numerically, validated. Third, replicate counts and statistical tests are not reported (triplicate plate readings over three days, but no n or significance analysis), preventing effect-size uncertainty assessment. Fourth, the model assumes equal mRNA and protein degradation rates and QSS ribosome binding β simplifications whose sensitivity is untested. The claim would be falsified if a constitutive circuit with faster protein turnover matched NAR's deviation under identical ribosome perturbation; that control is absent.
Bottom line: a coherent theory-experiment loop with a clean headline result (~20-fold vs ~1.5-fold), but modest scope (one strain, two perturbation modes, no statistical rigor, no bound-level validation). Confidence in the qualitative conclusion: moderate-high; in the quantitative bounds: low-moderate.
