Using ¹⁵N-NH₄⁺ tracer incubations (n = 4 per treatment) at Schiacchetiello inlet (Tyrrhenian Sea), the authors measured significant potential nitrification rates (PNR) only when the sponge C. nucula was present—alone or associated with P. oceanica; seagrass alone showed PNR close to zero . Rates were consistently higher in the dark (74–305%) and one order of magnitude higher in autumn (senescence) than spring (growth)—counter to ambient NH₄⁺ patterns (12.74 µM spring vs 2.76 µM autumn), which the authors attribute to spring competition for NH₄⁺ by the growing plant. 16S rRNA sequencing identified nitrifying taxa in the sponge microbiome: NOB Nitrospira (Nitrospiraceae, 2.62 ± 0.76%), AOB genera Cm1-21 and AqS1 (Nitrosococcaceae, 0.42 ± 0.12%), and AOA Candidatus Nitrosopumilus—with nitrifier families significantly enriched in sponge vs water column communities .
The sponge released NO₃⁻ (17.78 ± 3.49 spring; 23.59 ± 2.89 autumn µmol g DW⁻¹ d⁻¹) and NH₄⁺ in spring, while the association showed net NH₄⁺ fluxes near zero—suggesting plant/epiphyte uptake of sponge-derived DIN. Conversely, the seagrass was a constant DOC source (140–185 µmol g DW⁻¹ d⁻¹), and the association showed net DOC uptake in autumn (−40.37 µmol g DW⁻¹ d⁻¹), consistent with the sponge feeding on plant-released DOM via the sponge loop . Sponge-mediated PNR could support 8.35% (spring) and 47.38% (autumn) of holobiont primary production N demand, calculated from O₂ budgets (photosynthetic quotient = 1) and holobiont C:N ratios (23.3 spring, 27.8 autumn) .
Verdict: A well-executed isotope-tracer study that convincingly shows sponge-hosted nitrifiers supply DIN to the seagrass holobiont, with honest treatment of controls and statistics. The mutualism interpretation remains partially speculative (sponge benefit inferred, not measured), and ecosystem-scale extrapolation is premature. Confidence: moderate—supported by direct measurements, tempered by small n, single-site design, and model-based N-demand estimates.
