This Hamburg-Eppendorf preprint combines spinning-disk live imaging, 3D-STED, and 3D-MINFLUX exchange-PAINT (5β10 nm precision) to follow hNAIP/hNLRC4 inflammasome activation in primary human monocyte-derived macrophages infected with Y. enterocolitica WAC(pT3SS) β an engineered strain expressing the T3SS but no Yop effectors, used because wild-type Yersinia blocks NLRC4 activation via Yops ().
Key mechanistic sequence (observed): (1) galectin-3 marks phagosome rupture before eGFP-hNAIP accumulation at the disrupted phagosome, while eGFP-NLRC4 was never detected at the phagosome but accumulated later in distant specks (); (2) in 22 macrophages from 3 donors, ~90% of specks were preceded by one (59%) or two (30%) ruptured phagosomes, with rupture-to-speck latency of 0β71 min and no correlation between speck distance (0β28 Β΅m) and formation time (r = 0.1458) (); (3) triple-staining showed clusters shifting from NLRC4/NLRP3 (~50% at 30 min) toward ASC/NLRC4/NLRP3 (~60% at 120 min), with no ASC/NLRC4-only clusters β implying ASC joins pre-existing NLRP3/NLRC4 clusters (); (4) MINFLUX resolved putative single NLRC4 discs (max diameter ~45 nm, consistent with the ~32 nm cryo-EM disc plus labeling offsets), numbering 35β249 per mature speck (~1.65-fold higher after z-range extrapolation), with only ~30% of hNAIP localizations within 55 nm of an NLRC4 agglomerate ().
Strengths: first in-cell enumeration of individual NLRC4 inflammasome discs and hNAIP content within specks; appropriate strain controls (WAC, WA-314); temporal order established by live imaging (galectin-3 β hNAIP β distant speck β DRAQ7 membrane permeation); the low hNAIP content of mature discs is independently consistent with in vitro cryo-EM data ().
Weaknesses and blind spots: (i) the central causal model β phagosome rupture β hNAIP sensing β speck β pyroptosis β rests on correlation and colocalization; no hNAIP, NLRC4, or galectin-3 knockdown/knockout perturbation was performed, so causality is inferred, not proven (author-reported limitation is absent; BGPT inference); (ii) the effector-deficient WAC(pT3SS) strain is an artificial activator β wild-type Yersinia suppresses these pathways, so physiological relevance to natural infection is untested (BGPT inference; the paper acknowledges Yops block NLRC4) (); (iii) sample sizes are small: 22 macrophages, 16 specks for formation kinetics, 7 specks at the 40-min timepoint, and unreported numbers of MINFLUX-imaged specks β the wide 35β249 disc range may partly reflect low n; (iv) the 55 nm colocalization threshold and 20 nm agglomeration cutoff are assumptions that could inflate or deflate hNAIP-in-disc estimates; (v) a competing 2026 study argues human NLRC4 can directly sense flagellin independently of hNAIP and that hNAIP dampens flagellin-driven responses () β compatible with the sparse hNAIP-in-disc finding, but suggesting the "hNAIP-first" model may not generalize to flagellin-bearing bacteria; (vi) the paper itself flags that MINFLUX z-range truncation forces a 1.65Γ extrapolation of disc counts, assuming specks are spherical and the imaged layer is central β an unverifiable geometric assumption.
What would falsify the model: showing hNAIP is not enriched at galectin-3-positive phagosomes, or that specks form without prior phagosome rupture in T3SS-infected hMDMs; conversely, loss-of-function of hNAIP abolishing speck formation would strengthen it (BGPT inference).
Confidence: the descriptive observations (timing, counts, colocalization percentages) are well supported within the studied system; the mechanistic interpretation is plausible but correlational. Generalization beyond this engineered strain and beyond macrophages from three donors is uncertain.
