Culp and Christensen used a quantitative RT-PCR assay targeting the E1^E4 splice junction to resolve a literature conflict: EM-based studies of BPV-1 suggested entry within 30–60 min, while post-attachment neutralization experiments suggested virions remain accessible for many hours . Peak E1^E4 expression occurred at 48–120 h post-infection depending on the cell line, and neutralizing MAbs (H11.H3, H11.B2, G131S11.G3, H6.C6) plus the microbicide PSS could block the bulk of contributing virions even 24–72 h after adsorption—implying a large "window of neutralization" and minimal capsid conformational change during prolonged surface residence . HPV-40 and CRPV showed comparable slow kinetics, suggesting the finding generalizes across papillomavirus types.
Lower Ct = higher transcript abundance. SV40-transformed keratinocytes (KH-SV, BO-SV) expressed HPV-11 transcripts far more poorly than A431, HaCaT, or COS-7 cells.
Strengths: The QRT-PCR readout measures a product of successfully delivered viral DNA, so it selectively tracks the infectious pathway—unlike EM or fluorescent microscopy, which cannot distinguish infectious from non-infectious internalization, a key confound in prior fast-entry studies . Control experiments (mock rinsing, medium exchange, 4 degrees C incubation, spent-medium transfer) were thoughtfully included.
Weaknesses: The m.o.i. of 150 particles/cell (with unknown infectious fraction) exceeds natural exposure; the transcription readout cannot distinguish slow entry from slow post-delivery expression; and the authors' alternative explanation—that pinocytosed antibody neutralizes virus inside endosomes—was not directly excluded . Later work complicated the slow-entry picture: HPV appears to bind heparan sulfate and initiate infection at the basement membrane with sequential conformational changes , and innate inhibitors such as alpha-defensin HD5 block a delayed L2 furin-cleavage step, consistent with an extended post-attachment vulnerability window —broadly corroborating a prolonged neutralizable phase, though for HPV16 pseudovirions rather than authentic HPV-11.
Confidence: Moderate-to-strong for the core claim within these cell-culture conditions; weaker for extrapolation to in vivo epithelium, which the authors acknowledge. What would change the conclusion: orthogonal single-particle visualization showing bulk internalization within minutes in the same near-confluent monolayers, or failure of late-added MAbs to neutralize any cell line tested.
