This cryoEM study (14 Γ resolution, 2,269 particles) shows DAF/CD55 binding CVB3-RD across the virion surface rather than inside the canyon, achieving near-full occupancy of all 60 sites and physically blocking the CAR binding site (). SCR2 dominates contact (1,400 Γ Β² vs 558 Γ Β² for SCR3 and 272 Γ Β² for SCR4; SCR1 zero), placing most DAF residues used for virus binding on the face opposite convertase interaction (). A related NMR structure of DAF CCP2-3 (PDB 1NWV) provided the best-fitting central fragment, consistent with the flexible interdomain architecture ().
Plaque-reduction assays (>90% reduction) showed soluble DAF blocks CVB3-RD on RD cells while soluble CAR blocks wild-type CVB3 on HeLa cells, supporting a model where the bound DAF sterically occludes the canyon, forcing receptor competition (). The CVB3 footprint is unrelated to echovirus DAF footprints, supporting convergent evolution of DAF usage. Importantly, the authors themselves flag key unknowns: whether DAF alone suffices for entry (without canyon-binding pocket-factor displacement) is unresolved, and the classic VP2 residues 2108/2151 do not map to the observed footprint, weakening the direct genotype-phenotype link ().
Confidence in the structural map is high; mechanistic claims about entry are interpretive. Remaining blindspots: DAF ectodomain was yeast-expressed, His-tagged, and GPI-anchorβless, so native membrane-context affinity may differ; occupancy inferred from density could be influenced by the tag; and only two cell lines/two strains were tested. Data (EMD-1412, EMD-1411; PDB 2QZD/2QZF/2QZH) are publicly deposited, aiding reproducibility. Falsification would require SCR2-contact mutants that bind DAF normally or DAF-bound virus entering RD cells with canyon access intact.
