Using a filtration-based screen against host cells deposited on filtersβnecessary because AOB do not form plaques on solid mediaβthe authors recovered 12 lysates from two Alberta wastewater plants and assembled 45 high-confidence dsDNA phage genomes infecting four AOB species, the largest cultivated AOB phage collection to date (). Host-range testing of mixed lysates against the four AOB hosts showed 16 of 42 genomes infecting more than one host; 10 crossed the Nitrosomonas/Nitrosospira genus boundary and one phage (rov1) infected all four hosts. Infection was inferred from early cessation of nitrite production relative to untreated controls, confirmed by phage-specific PCR ().
Hierarchical clustering (GRAViTy v2, ViPTree, vContact2, VIRIDIC) organized the genomes into 21 family-level clades; six new families are proposed (Araviridae, Druantiviridae, Cernuviridae, Sinaviridae, Fulfuniviridae, Nantoviridae) with 34 genera. Eleven genomes are putatively temperate (integrase-positive), differing significantly from lytic genomes in %GC (p<0.0001) and size (p=0.0117). Genome sizes span 12.7 kb (ripi7_b) to a 338.7 kb jumbo phage (roc1). Structure-guided annotation (Phold) revealed AMGs including a complete ABC transporter cassetteβclaimed as the first in any phage genomeβiron/heme acquisition systems, glycolysis and pentose phosphate pathway enzymes, plus ARGs and paired defense/anti-defense (CBASS/Pycsar/NARP) systems ().
Metavirome screening is a striking result: relatives of only six phages (ripk1, ripk2, ripk4, roc1, roc2, rog2) were found across 478 soil viromes, and 676 wastewater viromes from 63 cities showed no substantial coverage of most genomesβsuggesting this cultivated diversity has been systematically missed by metagenomics (). This converges with independent freshwater evidence of amoC-encoding phages actively modulating nitrification, reinforcing that AOM viruses are ecologically significant ().
What would weaken the conclusions: purified single-phage isolates failing to reproduce nitrite-cessation phenotypes, or re-analysis showing genome assemblies derive from host contamination. Confidence in the taxonomic/genomic claims is high; confidence in functional and ecological claims is moderate pending wet-lab validation.
