Gutmann et al. pooled a 1573-gRNA CRISPRi library (dCas9-Mxi1, ATc-inducible) targeting 161 transcription factors and 129 protein kinases in diploid S. cerevisiae BY4743, then ran 25-generation competitive screens in synthetic complete medium (SCM), SCM + 10% spruce hydrolysate, and SCM + 45% inhibitor cocktail (furfural, 5-HMF, formaldehyde, acetic acid, formic acid, vanillin, coniferyl aldehyde, p-hydroxybenzaldehyde), each in triplicate . QC is solid: essential genes were depleted as positive controls, and guide-level fold changes correlated with an independent prior screen (Pearson RΒ² = 0.67, p < 2.2e-16) .
Twenty-four genes showed significant hydrolysate fitness effects (FDR < 0.05, β₯2 gRNAs with |log2FC| β₯ 1). Seven repressions reduced hydrolysate fitness (YAP1, HAA1, HOG1, PBS2, UGA3, CDC15, UME6); three increased it (DOT6, SKO1, BUB1). Five hits (dot6Ξ, sko1Ξ increased; hog1Ξ, pbs2Ξ, stb5Ξ decreased fitness) were independently recapitulated with BY4741 deletion strains . Comparing hydrolysate and inhibitor-cocktail screens showed that roughly one-third of hydrolysate-specific functions (BUB1, DOT6, UME6, YAP1) were reproduced by the defined inhibitor cocktail, indicating toxicity explains at least that fraction, a lower bound the authors flag . Notably, HAA1, HOG1, PBS2, SKO1, and UGA3 effects did not appear with the cocktail, implying osmotic and pH stressβnot only the eight toxinsβdrive much of the hydrolysate phenotype.
Effect-direction schematic; only the HOG1 guide log2FC (-3.35) and SKO1/HOG1 magnitudes were numerically reported, so per-gene bar heights beyond HOG1 are schematic (BGPT inference from hit classification, not quantitative replication).
Confidence: high for the listed hit genes as screen-level observations; moderate for mechanistic interpretation (HOG pathway, Sko1 de-repression) which rests on prior literature plus spot-plating of one guide; unknown whether effects replicate in industrial hydrolysate feedstocks beyond SEKAB spruce material.
