In developing Arabidopsis first rosette leaves, DR5::GUS auxin-response domains reliably precede procambial differentiation: a distal focus (DF) of subepidermal DR5 expression appears before the midvein forms, incipient secondary/tertiary vein DR5 stripes emerge before oriented cell divisions produce narrow procambial files, and DR5 expression disappears once vessel elements differentiate .
Under auxin-transport inhibition (NPA 0.1β10 Β΅M), DR5 expression shifts toward the leaf margin in a dose-dependent manner β mirroring the previously reported margin-restricted vascularization under NPA (Mattsson et al., 1999; Sieburth, 1999). This concordance under perturbation supports transported-auxin accumulation, not local synthesis or spurious DR5 activation, as the patterning mechanism .
MONOPTEROS (MP/ARF5) is a limiting factor in auxin signal transduction: mp mutants show severe auxin insensitivity in cotyledon-expansion and adventitious-rooting assays (94% of wild-type cotyledons rooted vs 25% axr1-12, 11% mp), and 35S::MP overexpression elevates IAA1, IAA19, AtHB8, and AtHB20 transcripts; auxin induction of IAA1/IAA19 is reduced in mp .
A new finding was the vascular expression profile of AtHB20 (class I HD-ZIP): diffuse around emerging veins early, restricted to veins and fascicular cambium in mature leaves, with expression strictly dependent on MP dosage though its rapid auxin inducibility fell below statistical significance due to high variability .
Confidence: High for the descriptive DR5-procambium correlation and MP auxin-insensitivity assays (large samples, replicated reporter patterns, two independent mp alleles); moderate for the MP-dependence of AtHB8/AtHB20 (reliant on overexpression, variable northern-blot signals). What would change the conclusions: direct auxin measurements showing DR5-independent auxin maxima at vein sites, or vascular patterning intact in auxin-response-defective backgrounds.
