Ben Arfa et al. tested five aroma compounds β carvacrol, eugenol, menthol, and two synthesized carvacrol derivatives (carvacrol methyl ether, carvacryl acetate) β against seven microorganisms using both broth (MIC) and microatmosphere vapour (MID) methods, with experiments in at least triplicate
MIC values from Table 2 (visualized below) show carvacrol consistently lowest; L. plantarum resisted all compounds. Notably, MICs were cidal, not merely static β subculturing after MIC exposure yielded no colonies
The authors propose a two-factor model: (1) sufficient hydrophobicity (logP β 3β4) allows membrane accumulation, and (2) a free hydroxyl group coupled to a delocalized electron system permits proton exchange, collapsing the proton motive force and depleting ATP β building on Ultee et al.'s hypothesis . However, the proton-exchange mechanism is inferred, not directly measured (no membrane potential or ATP assays were performed here).
Subsequent work confirms the structureβactivity logic: carvacrol MICs of 128 mg/l against erythromycin-resistant Group A Streptococci with synergy (FIC β 0.25) with erythromycin , and membrane disruption confirmed by propidium iodide uptake in Xanthomonas (carvacrol MIC 0.25 mg/ml) . A Phase 2/3 trial of carvacrol/isothymol against SARS-CoV-2 pneumonia was completed but results quality cannot be judged from registration data alone .
Confidence: Moderate β the descriptive SAR findings are internally consistent and corroborated by later membrane studies, but the mechanistic claim rests on inference, and strain coverage is narrow.
