This study investigates the antigen specificity and clonal architecture of CD8+ T cells derived from the cerebrospinal fluid (CSF) and peripheral blood of multiple sclerosis (MS) patients. Leveraging single-cell RNA sequencing and T cell receptor sequencing (TCR-seq), the authors uncouple the complexity of immune activation in MS by identifying highly expanded clonotypes that preferentially target Epstein-Barr virus (EBV) epitopes and novel mimotopes.
The authors acknowledge a limited sample size, particularly in the control group, which may affect the wider applicability of the findings. Future studies involving larger and more diverse cohorts are needed to validate the pathogenic roles of these CD8+ T cells and to explore if similar antigen-specific responses occur across different MS subtypes
This paper provides a compelling analysis linking EBV-specific CD8+ T cells with MS. The integration of high-resolution single-cell approaches establishes a foundation for future biomarker discovery and targeted immunotherapies in multiple sclerosis, although additional validation is essential.
