The assay couples G6PD (from Leuconostoc mesenteroides) oxidation of G6P to a NADP+/NADPH recycling reaction that reduces WST-1 to a water-soluble yellow formazan via 1-mPMS, read at 440 nm in 96-well format . The recycling design amplifies signal so NADP+ (25 µM) works far below the stoichiometric ~200 µM used in classic assays, driving LOD to 0.15 µM and LLOQ to 0.5 µM — a clear sensitivity gain over direct NADPH measurement at 339 nm . Specificity was tested against F6P, G1P, ribulose-5P, ribose-5P (all 100 µM) and glucose (100 mM); only ribulose-5P showed minimal interference.
Calibration in deproteinized, heat-inactivated FBS gave a reproducible curve (slope 0.001169, n=8), spike recoveries of 91.4–92.2% at 30–60 µM spikes, and measured FBS G6P of 6.2 ± 0.5 µM, cross-checked against a conventional enzymatic assay (5.9 ± 0.3 µM) . This orthogonal agreement is the paper's strongest credibility signal. BGPT inference: the record's legacy extraction of "56.7–63.2% recovery" appears to confuse found (absolute) values with recovery percentages; the paper itself reports ~92% recovery, and the found-vs-expected values align.
At the LLOQ (0.5 µM), within-run and between-run %CV were 13.2% and 15.9% — within the 20% bioanalytical acceptance criterion but close to the margin, meaning low-end quantitation carries real uncertainty . Key blindspots: (1) only FBS was validated — human serum, hemolysates, and tissue extracts with endogenous G6PD or redox-active compounds were not tested, and 1-mPMS is redox-active so reducing agents in untested matrices could cause background; (2) ribulose-5P interference is acknowledged but not characterized quantitatively at low G6P levels; (3) ribulose-5P is itself a downstream product of the G6PD reaction, so coupled-pathway samples warrant caution; (4) no independent cross-lab reproducibility; (5) WST-8 substitution was mentioned but not validated because of kit-only availability — a vendor-dependent limitation. A 1997 PMS/MTS tetrazolium assay for the same metabolites already existed , so the novelty lies mainly in the WST-1/recycling 96-well implementation and serum validation rather than concept. The core claim is well supported; generalizability beyond FBS remains unproven. Confidence: moderate-high for reported performance; lower for broader applicability.
