Costache, Cailleau, Fernandez-Mateos, Oriol, and Mollicone (INSERM U178, Paris-Sud) synthesized the then-complete catalogue of human fucosyltransferase genetics: seven functional genes (FUT1–FUT7) plus the Sec1 pseudogene, organized into a 19q13.3 cluster (FUT1–FUT2–Sec1, spanning ~100 kb with FUT1–FUT2 at 35 kb distance) and a 19p13.3 cluster (FUT6–FUT3–FUT5, ~50 kb), with monomorphic FUT4 on 11q21 and FUT7 on 9q34.3 . High within-cluster sequence identity (~70–90%) versus low cross-family identity (<20%) supported duplication-translocation-divergent evolution .
The paper's phylogenetic tree (28 coding sequences, PHYLIP Fitch-Margoliash) dated the FUT1/FUT2 split before the great mammalian radiation (~80 MYA), placed Lewis/myeloid/leukocyte subfamily divergence before the bird-mammal split, and predicted lower mammals lack separate FUT3/FUT5/FUT6 orthologs—later confirmed by chimpanzee having three separate Lewis-branch genes and bovine having a single ancestral-like Lewis gene .
The developmental-switch analogy with hemoglobin genes (ubiquitous embryonic FUT4/LeX in 5–10 week embryos switching to FUT7/FUT6/FUT3 in differentiated tissues, derepressing telomere-to-centromere within clusters) was a notable, testable conceptual claim .
Population genetics was a genuine strength: race-specific inactivating alleles (Caucasian FUT2 G428-A nonsecretor; Oriental/Polynesian C571-T, C628-T, G849-A; A385-T weak secretor in ~20% of Orientals; Indonesian Lewis T1067-A) implied these mutations arose very recently in evolution, in sharp contrast to ancient ABO trans-species alleles .
The proposed two-domain α/β fold inspired by bacteriophage T4 β-glucosyltransferase, with GDP-fucose binding between domains and conserved Asp/Glu donor-recognition residues (Glu247 of FUT6), was based on HCA and threading—purely computational . Subsequent work largely vindicated the family-level framework: the 2025 Nature Communications review confirms a GT-B fold with two Rossmann-like domains and distinct Clade 1 (FUT1/2/8, POFUT1/2) versus Clade 2 (FUT3–7, FUT9, POFUT3/4) organization, though catalytic mechanisms are enzyme-specific (e.g., Glu-based catalytic bases, no obligatory metal cofactors) rather than the single unified model sketched here . The hypervariable stem-domain specificity claim was later mechanistically pinned down: a single residue (Trp111 in FUT3) determines type 1 versus type 2 acceptor transfer, and aromatic residues permit α1,4 activity .
What would change this conclusion: definitive absence of the proposed cluster organization in modern genome assemblies, failure of W111/stem-domain mutants to alter acceptor specificity, or phylogenies (maximum-likelihood, as later used for bovine FUT7 with >98% bootstrap support) contradicting the pre-mammalian-radiation H/Se split .
