Durkin, Gao, and Nachman cross-fostered wild-derived inbred house mice from temperate (SARA, NY) and tropical (MANA, Brazil) localities within 24h of birth, weighing pups at PND0β21 . SARA-CF pups (n=25) were significantly lighter than non-CF SARA (n=43) during nursing (p<0.0001), while MANA pups showed no cross-fostering effect; weights converged by weaning .
Maternal DEGs showed significantly higher trans-regulation than genetic DEGs (Fisher's exact p<0.05 both reciprocal crosses); cis-only genes were over twice as numerous as trans-only among genetic DEGs . A 66-gene WGCNA module (r=0.88, p<0.0001) linked to maternal status was enriched for cholesterol biosynthesis (Hmgcs1, Mvd, Sqle), RA metabolism, and zinc/detox genes .
Rearing dam explained 45% of pup microbiome variance (strain 13%, PERMANOVA p<0.01); Lactobacillus abundant in SARA-reared, absent in MANA-reared pups . Intersection of QTL LOD regions, MP body-size genes, cis-DEGs, and PBSn1 outliers yielded 32 candidates including Slc2a2, Evc, Evc2, Igfbp7, Eif2ak4 (permutation p<0.01) .
No direct milk output/composition measurementβtime-in-nest served as nursing proxy; only two inbred strains; liver-only transcriptomics; modest microbiome n (4/group) . Data/code at NCBI PRJNA1480043/1482167 and GitHub .
