The reviewed papers reveal a laboratory with exceptional depth in papillomavirus-host interactions, built around two landmark mechanistic threads. The first is the E6-p53 axis. Howley's group purified E6-AP from 270 confluent Saos-2 plates through a seven-column chromatography scheme, sequenced peptides, cloned the 865-aa cDNA (predicted mass 99,289 Da), and reconstituted E6-mediated p53 degradation in vitro with E6-AP and ubiquitin β an achievement that first linked a viral oncoprotein to the ubiquitin-proteasome system and prefigured the entire HECT-E3 field . Follow-up genomic work mapped UBE3A splicing, identifying five mRNA subtypes encoding three isoforms (98.9 and 101.7 kDa characterized), connecting the locus to Angelman syndrome .
The second thread centers on E2 and chromatin tethering. Using TAP-MS, Howley's lab identified Brd4 as the major BPV-1 E2 partner, mapped binding to Brd4's C-terminal domain (aa 1047β1362), and showed dominant-negative Brd4-CTD disrupts mitotic chromosome localization and reduces transformation in C127 cells . Quantitative half-life work later showed Brd4-CTD extends BPV1 E2 half-life from 0.51 h to >9 h and HPV16 E2 from 0.78 h to >6 h by blocking a Cul3-based E3 ubiquitylation axis (HPV16 E2: 1.7 h β 6.5 h with DN-CUL3) :
Reported values . The >h bars are lower bounds, plotted at their reported floors.
Howley also charted the E2 domain architecture (C-terminal dimerization; N-terminal activation), with deletion of aa 1β195 reducing transactivation to 7% of wild-type and mixed translation producing 1:2:1 heterodimer patterns , and extended the ubiquitin theme to E6 itself, showing USP15 extends HPV16 E6 half-life from 68 to ~278 min in a catalytically dependent manner . Earlier p53 domain-mapping work (mutants 179/248/273 in H358 cells; 179 nearly fully transdominant; p53A43 retaining 30β70% transformation suppression in rat embryo fibroblasts across six experiments) anticipated the oligomerization-dependent dominant-negative model , and PCNA-Fen1/p21 competition work (Kd ~15 vs ~60 nM; SPR stoichiometry ~2.47 Fen1 per trimer) showed breadth beyond virology .
Recurring strengths: mechanistic completeness (purification β cloning β reconstitution β in-cell validation), orthogonal assays (co-IP, ChIP, microscopy, CHX chase, SPR), and reproducibility (independent replicate experiments consistently reported). Recurring weaknesses, acknowledged in the papers themselves: heavy reliance on overexpression in single cell lines (C33A, CV-1), incomplete identification of the Cul3 BTB adaptor , and Fen1-PCNA stoichiometry ambiguity (2 vs 3 per trimer) . Funding disclosures (NCI R01 CA77385, P01 CA50661, R01 CA050661, R37 CA64888) indicate sustained competitive NIH support with no declared conflicts of interest . OpenAlex author metrics were unavailable (API rate-limited), so citation totals could not be independently verified here. What would weaken this assessment: failure of the E6-AP or Brd4-tethering models in physiological infection contexts β none reported in the reviewed evidence; the mechanistic cores have proven durable.
