John Kuriyan's OpenAlex profile records 512 works with 51,416 citations and an h-index of 111, with sustained productivity from 1986 through 2026 and roughly 1,500β2,200 citations per year over the last decade. His most-cited works include the molecular dynamics crystallographic refinement method with BrΓΌnger and Karplus (1,956 citations), the STI-571/Abl kinase structure (1,785), the "Conformational Plasticity of Protein Kinases" review (1,761), and the EGFR allosteric activation paper (1,650). Topic fingerprints span biology, chemistry, cell biology, and biochemistry, consistent with a structural-mechanistic research program rather than a narrow specialty.
The EGFR allosteric activation study (Cell, 2006) is a model of convergent mechanistic evidence: crystal structures of the autoinhibited kinase domain (PDB 2GS2/2GS6/2GS7), vesicle-based reconstitution showing concentration-dependent activation, the L834R mutant with ~20-fold higher kcat/KM, and cell-based confirmation that dimer-interface mutations impair autophosphorylation at Tyr1045/1068/1173. Reported limitationsβtruncated constructs, overexpression assays, and possible lattice artifactsβare honestly scoped, and the paper scores 9/9/8 on quality/novelty/generality in extraction. The work defined the asymmetric CDK/cyclin-like dimer mechanism now foundational to ErbB biology.
The CaMKII linker-length study (eLife, 2020) demonstrates continued mechanistic leadership decades into the career: single-molecule TIRF imaging of tens of thousands of holoenzymes showed that short kinase-hub linkers favor activating Thr286 phosphorylation while long linkers bias inhibitory Thr305/306 (CaMKII-Ξ² ~95% vs CaMKII-Ξ± ~30% pThr305/306), supported by heterooligomer co-expression (~80% colocalization), phosphatase assays, and a kinetic model. Notably, Kuriyan is disclosed as a Senior Editor at eLife, a declared non-financial editorial role worth noting for transparency.
The 2026 ClpXP mechanochemistry preprint extends the program into AAA+ motor energetics with optical tweezers and cryo-EM across seven engineered hexamers (N=47β69 traces each). Rigidity of the central coupler tracks directly with coupling efficiency: WT uses ~2 ATP/step with 75% GFP unfolding probability, whereas the alternated WMWMWM mutant consumes ~7 ATP/step with 0% unfolding and F1/2 dropping from 19 to 13.5 pN. The authors transparently flag that the transient Class-II pre-power-stroke state is inferred from static snapshots under ATPΞ³S.
The 2026 protease-resistant HR2 fusion inhibitors study shows translational reach: deep-mutational-scan-guided design yielded HR2-NHN, resistant to TMPRSS2 cleavage, retaining IC50 1.9 nM after 72 h pre-incubation in Calu-3 cells (vs wildtype shifting 2.6β8.9 nM), and an 11 h vs 8.45 h apparent antiviral half-life in BALB/c mice (n=4β5/group, bootstrap AUC significant). Stated blindspotsβfemale mice only, one variant in vivo, no pharmacokinetics, both peptides inactive after 24 h pre-incubation in A549-AT cellsβare candidly reported.
Strengths: mechanistic clarity, integration of structure with quantitative biochemistry, and unusually explicit falsification criteria and limitations across the sampled corpus (paper quality scores 8β9 throughout). Weaknesses: nearly all highlighted work is in vitro or overexpression-based, so physiological extrapolation is inferential; several 2026 records lack complete conflict-of-interest statements; the ClpXP and HR2 papers have zero recorded incoming citations (very recent), so field uptake is not yet assessable; and the CaMKII kinetic model is a two-dimer simplification of a 12β14 subunit holoenzyme. What would change the assessment: failed replication of the asymmetric dimer mechanism in native membranes, or in vivo falsification of the linker-length model in neurons. Confidence: high on metrics and reported findings; moderate on translational impact of 2026 work given its recency.
